Technical background

As a key bioactive molecule, long peptides are widely used in drug development, vaccine design and biomedical research. The technology of synthesizing growth peptides has always been one of the hot topics in the field of biomedicine.

Long peptide synthesis technology plays an important role in biomedical research, aiming to improve the synthesis efficiency, reduce the cost, and ensure the quality and purity of the product.

In the study of protein chemical synthesis, solid phase synthesis is the most popular and convenient method. This method is also effective in the synthesis of long chain polypeptides.


Technical principle

At present, the following methods are commonly used to solve the problems existing in the synthesis of long peptides:

(1) Microwave synthesis: Microwave synthesis is a method of using microwave radiation to accelerate chemical reactions. Microwaves are electromagnetic waves with frequencies in the range 0.3-300 GHz. Microwave radiation can be converted into internal thermal energy by electromagnetic radiation inside the molecule, thus increasing the reaction rate. Under microwave radiation, the vibration and rotation rate of molecules increase, making chemical reactions occur faster. For some amino acids which are difficult to condense in the synthesis process, we use microwave method to synthesize them. The effect is remarkable, and the reaction time is greatly shortened, while the production of two main by-products is reduced.

(2) fragment synthesis method: When some peptides are difficult to synthesize by conventional synthesis methods and difficult to purify, we will also use a certain number of amino acids in a certain segment of the peptide to condense into the peptide chain as a whole, which can also solve many problems in synthesis.

(3) hydrazide synthesis method: The method of synthesizing polypeptides by hydrazide is a method of chemically selective reaction between N-terminal Cys polypeptide synthesized in solid phase and C-terminal polypeptidyl hydrazine to form an amide bond and realize the polypeptide connection. According to the location of Cys in the peptide chain, the whole peptide chain is divided into multiple sequences and synthesized respectively. Finally, the target peptide is obtained through liquid phase condensation reaction. This method not only greatly reduces the synthesis time of long peptide, but also significantly improves the purity of the final product, so it is widely applicable to the synthesis of long chain polypeptides containing Cys.
